MAJOR
RESEARCH INTERESTS:
My laboratory studies the
underlying pathophysiology of human male infertility with the long-term goals
of developing new diagnostics and new treatment options.
Study of human testis
biopsies from infertile men with varicocele, with hypospermatogenesis without
varicocele, with Sertoli cell only syndrome and with maturation arrest
unexpectedly indicated that testicular accumulation of heavy and transition
metal ions and an alteration in calcium homeostasis are common intermediaries
in these syndromes which result in decreased sperm densities in the ejaculate.
Calcium signaling is a very important regulator of testicular germ cell
apoptosis, which in turn controls sperm production. The heavy and transition
metal ions, which are calcium competitors, may be interfering with calcium
signaling. Ongoing studies employ a combination of epidemiology, trace metal
analysis, pharmacology, gene expression profiling via DNA microarrays and
proteomics with human testis biopsies and with testes from rats exposed to lead
or cadmium. These studies are being performed in order to identify the calcium
signaling pathways disrupted in metal-induced hypospermatogenesis and the
initiating event, which is likely to be different in each syndrome. Important
findings so far include roles for ion channel expression modulation and for ion
channel alternate splicing as protective responses and determinants of
susceptibility or resistance to heavy metal-induced apoptosis, and for maternal
smoking as a primary effector of low sperm counts in male progeny.
