多激酶抑制剂索拉非尼可诱导前列腺癌PC-3细胞发生caspase依赖的凋亡

本研究观察多激酶抑制剂索拉非尼(sorafenib)对前列腺激素非依赖肿瘤细胞存活的影响及其作用的信号途径。以人前列腺激素非依赖PC-3肿瘤细胞为研究对象,索拉非尼在抑制其细胞外信号调节激酶(extracellular signal regulated protein kinases, ERK)磷酸化的浓度下,可使PC-3细胞线粒体膜电位下降,细胞凋亡。索拉非尼使PC-3肿瘤细胞内的髓细胞白血病基因-1(myeloid cell leukemia-1, MCL-1),存活素(survivin)和细胞凋亡抑制蛋白2(cellular inhibitor of apoptosis protein 2, c-IAP2)表达下调,并使PC-3肿瘤细胞内的caspase-3裂解,细胞色素(cytochrome c)释放,但未检测到凋亡诱导因子(apoptosis inducing factor, AIF) 从线粒体转位到细胞核,且广谱caspase抑制剂z-VAD-FMK基本完全抑制sorafenib所诱导的凋亡。综上,sorafenib可下调前列腺癌激素非依赖细胞内的抗凋亡蛋白表达并可诱导caspase依赖的细胞凋亡。
