敲减CatSper2表达实现对雄性大鼠受精能力的调控

随着不同离子通道亚基被定位在精子上,其作为避孕靶标受到越来越多的关注。选择性敲减离子通道的亚基能够成功实现避孕目的,并且不会产生副作用。本研究应用活体电转结合睾丸网微注射介导质粒转染大鼠睾丸,以期通过阻断精子超活化通路CatSper2蛋白表达,实现对雄性大鼠生殖调控的目的;结果表明,大鼠睾丸网质粒微注射后电转处理在体内能够获得较高的转染效率和对CatSper2蛋白表达的良好敲减;随着CatSper2 蛋白表达被阻断,处理组大鼠无论是附睾精子超活化率、体外受精率、粘滞性溶液中迁移率,还是胞内Ca2+峰等指标均显著低于对照组大鼠(P<0.05),并且这种低超活化率和低受精率的状态能够持续60天之久;同时,通过对附睾精子体外存活能力、睾丸组织学等分析证实,活体电转和睾丸网注射的处理并不会对大鼠睾丸功能、精子发生及精子活动性等指标产生显著的影响;这些结果的证实,印证了我们阻断精子超活化通路中关键蛋白CatSper2表达,实现雄性避孕目的的思路是可行。
