评价小鼠附睾头部和尾部精子DNA完整性的4种方法的比较

众所周知,精子穿过附睾的主要变化之一是精子染色质压缩,到达附睾头部的精子染色质处于完全压缩状态。本文的研究目的是比较末端标记法(TUNEL)、彗星试验(comet)、精子染色质结构试验(SCSA)和精子染色质扩散试验(SCD)这四种方法鉴别附睾头部和尾部处于不同成熟阶段(染色质压缩和DNA片段化)的精子的能力。结果表明,未成熟精子的DNA片段可用comet和SCSA检测到, SCD或TUNEL检测不到。
