siRNA-HMGN5通过影响线粒体相关的信号通路及调节Bcl-2家族蛋白引起前列腺癌细胞系凋亡

HMGN5作为核小体结合蛋白,能够影响染色质的解聚及调控基因的转录。我们采用慢病毒介导的siRNA-HMGN5转染前列腺癌LNCaP细胞系后发现细胞凋亡增加,因此我们进一步针对其中的作用机制展开研究。前列腺癌雄激素依赖细胞系LNCaP转染siRNA-HMGN5后,用Annexin V-PE/7-AAD染色法及TUNEL染色法检测细胞凋亡,JC-1染色法测定线粒体膜电位的变化,real-time PCR检测HMGN5及GAPDH的mRNA表达情况,western blot检测Bcl-2家族及其它凋亡相关蛋白的表达情况,并检测细胞裂解液中caspase的活性。PCR及western blot结果证实转染siRNA-HMGN5可有效地降低HMGN5的表达。HMGN5表达的下调引起LNCaP细胞凋亡的增加,线粒体膜电位的下降,Bax及Bax/Bcl-2比值的升高及caspase3的活化。因此我们认为siRNA-HMGN5能够有效的通过影响线粒体相关的信号通路及调节Bcl-2家族蛋白引起前列腺癌细胞系LNCaP凋亡,HMGN5可作为潜在的治疗前列腺癌的靶向分子。
