Delta Np63基因沉默可上调膀胱肿瘤细胞株5637的细胞粘附能力

有研究表明delta Np63表达缺失可能与肿瘤的恶性行为有关。相反也有研究认为delta Np63可能是一种癌基因。Delta Np63的高表达可在鳞状上皮肿瘤和膀胱肿瘤中检测到。本文研究了delta Np63基因沉默,对膀胱移行细胞癌细胞粘附能力的影响和调控。5637细胞稳定转染delta Np63 shRNA特异性表达质粒。免疫荧光检测转染效率。检测5637细胞转染delta Np63 shRNA特异性表达质粒后与血管内皮细胞的粘附能力。激光共聚焦显微镜检测F-actin和细胞骨架的改变。流式定量检测F-actin的改变。transwell小室检测细胞侵袭能力的改变。Delta Np63 shRNA基因沉默后,5637细胞与血管内皮细胞的粘附能力增强,F-actin的表达增加,细胞侵袭能力下降。Delta Np63基因沉默后,可能通过上调F-actin的表达来增强5637细胞与血管内皮细胞的粘附能力,从而阻止膀胱肿瘤的侵袭和转移。
