冷诱导RNA结合蛋白(CIRP)的下调可以激活有丝分裂原激活蛋白激酶(MAPK)并且削弱小鼠睾丸的生精功能

冷诱导RNA结合蛋白(CIRP)在正常睾丸中表达,而在隐睾、精索静脉曲张和环境高温等受热刺激的睾丸中表达量明显下降。本研究的目的是探讨CIRP在睾丸中的生理功能。我们在睾丸中利用RNA干扰技术成功下调CIRP的表达后,用HE染色法观察生精小管形态学变化,TUNEL法观察生精细胞的凋亡情况,Western blotting 检测有丝分裂原激活蛋白激酶(MAPK)信号通路的相关蛋白表达的变化。我们发现,在睾丸中用siRNA干扰技术下调CIRP蛋白的表达是一种可行的方法。和对照组相比较,siRNA干扰组平均生精小管直径、生精细胞层数明显减少,生精小管凋亡率明显增加,p44/p42,p38和SAPK/JNK MAPK信号通路被激活。总而言之,在小鼠睾丸中下调CIRP的表达会导致生精细胞凋亡,可能的机制是激活p44/p42,p38和SAPK/JNK MAPK信号通路途径,为进一步研究热应激所致的生精功能损害的机制提供理论基础。
