转染VEGF165的内皮祖细胞移植恢复糖尿病ED大鼠的勃起功能

探讨血管内皮细胞生长因子(VEGF165 )基因转染大鼠骨髓内皮祖细胞(EPCs )移植到糖尿病性勃起功能障碍(DMED )大鼠阴茎组织后,观察其对勃起功能的影响。腹腔注射链脲霉素(STZ )建立DMED 大鼠模型,成模后将基因修饰的EPCs 分别移植于三组DMED 大鼠阴茎组织。结果发现:海绵体内压(intracavernosal pressure,ICP )检测结果表明,实验组大鼠的基础ICP 和电刺激诱导ICP 比空白对照组及控制组明显增高(P<0.01 )。组织学检查发现在实验组大鼠阴茎海绵体中有大量血管增生。荧光显微镜观察提示大量移植的EPCs 在实验组中存活,分化为内皮细胞,并整合到大鼠阴茎新生血管中。基于以上结果,我们认为转染VEGF165 基因修饰的EPCs 能有效改善DMED 大鼠勃起功能。
