PSP94或CRISP-3调节的生长抑制具有前列腺癌细胞株特异性

已有研究证明人类精浆中前列腺分泌蛋白94(PSP94)和富含半胱氨酸分泌蛋白3(CRISP-3)相互作用。有趣的是,PSP94表达在大多数前列腺肿瘤中降低或消失,而CRISP-3表达在前列腺癌中与正常前列腺组织相比却上调。为更好地了解这两种蛋白各自对于前列腺肿瘤形成的作用及其相互作用在功能方面的相关性,我们使PSP94和CRISP-3各自以及两者共同异位整合表达在三种前列腺细胞系上(PC3、WPE1-NB26、LNCaP),并进行生长抑制检测。用RT-PCR和Western blot测定PSP94和CRISP-3的表达,构建了PSP94和CRISP-3的哺乳动物表达,对重组蛋白的表达、定位和分泌进行转染分析,然后用Western blot和免疫荧光检测。用克隆形成细胞活存率实验和转染检测PSP94和CRISP-3的异位整合表达对细胞生长的作用。研究结果表明PSP94和CRISP-3都能引起细胞系特异性的生长抑制。虽然CRISP-3阳性细胞系会被PSP94抑制,但无论PSP94是否存在,CRISP-3调节的生长抑制都不受影响。这表明CRISP-3参与了前列腺肿瘤形成中非PSP94依赖的活动。
