韩国良性前列腺增生病人的前列腺体积与其一氧化氮合成酶2基因多态性的关联

良性前列腺增生(BPH)的确切病因仍不清楚,但我们知道免疫炎症过程在BPH发生和发展的发病机制中起一定作用。一氧化氮合成酶2(NOS2)诱导的表达与前列腺癌和BPH等前列腺疾病密切相关。本研究的目的是探讨NOS2多态性与BPH的关系。我们对205名对照受试者和229名BPH病人采用直接测序和限制性片段长度多态性分析,在NOS2基因中测出三种单核苷酸多态性(SNPs),包括rs2779248(启动子, –278 T/C)、rs10459953(5'-非翻译区)和rs2297518(外显子16,错义,Ser608Leu)。比较BPH病人和对照受试者的基因型和等位基因频率,并对其与BPH病人之间的关联进行分析。用SNPStats、SNPAnalyzer、HelixTree程序分析SNPs。SNPs与对照组和BPH病人分组无相关性。在BPH组内部,SNPs与PSA水平高低有相关性,但有一个SNP与BPH病人的前列腺体积相关(rs10459953,在共显性模型中,OR = 0.44,95%CI = 0.29–0.65,P < 0.0001;在显性模型中,OR = 0.23, 95% CI = 0.12–0.46, P < 0.0001; 在隐性模型中,OR = 0.46, 95% CI = 0.24–0.86, P = 0.015)。我们发现NOS2 SNP(rs10459953)的基因序列与前列腺体积大小有很大的相关性。这一结果提示NOS2可能与BPH病人的前列腺体积相关。
