一种新型抗凋亡蛋白抑制剂Apogossypolone体外诱导前列腺癌PC-3与LNCaP细胞的自噬

目前对于侵略性前列腺癌还没有有效的治疗方法,据报道棉酚对许多类型的肿瘤细胞有潜在的抗肿瘤活性。它可以增强肿瘤细胞对烷化剂的敏感性、消除多药耐药及肿瘤的转移,但是它是否能诱导自噬还不明确,为此我们研究了Apogossypolone(ApoG2)及左旋棉酚在体外对人前列腺癌细胞系PC-3与LNCaP的抗增殖活性。用ApoG2处理PC-3及LNCaP 细胞后出现自噬的几个典型特征,包括胞浆内膜状液泡及酸性囊泡细胞器、自噬相关LC3-II与 beclin-1 蛋白表达增强,使用3-甲基腺嘌呤抑制自噬可以促进二个细胞系的凋亡。总之,该研究表明对于前列腺癌细胞,自噬的诱导作为一种防御机制可以拮抗凋亡。
