下调NSBP1基因能抑制前列腺癌细胞在体内体外的生长

本研究构建了人核小体结合蛋白1(NSBP1)基因RNA干扰慢病毒表达载体,探讨了NSBP1基因与前列腺癌DU145细胞G2/M期阻滞和凋亡之间的作用,并且检测抑制NSBP1蛋白表达对DU145细胞体内体外生长的影响。在DU145细胞中抑制NSBP1表达,MTT法检测细胞的生长比率,流式细胞技术检测细胞G2/M期和凋亡情况。检测裸鼠种植肿瘤的生长情况,分别应用RT-PCR和Western blot检测NSBP1、CyclinB1及Bcl2基因在体内体外的mRNA和蛋白表达。我们发现抑制NSBP1蛋白表达后,细胞的生长比率下降了22.6%,裸鼠移植瘤生长减慢,G2/M期细胞循环被阻滞8.78%,同时凋亡细胞数是阴性对照组的2.19倍。并发现CyclinB1和Bcl2的mRNA和蛋白表达水平下降。抑制NSBP1表达能在体内体外抑制DU145细胞生长,部分原因是NSBP1抑制诱导了G2/M期阻滞和细胞凋亡。我们的数据说明抑制NSBP1表达引起G2/M期阻滞和凋亡或许是因为NSBP1下调CyclinB1和Bcl2表达的结果。
