瞬时受体电位通道C6在肝细胞生长因子诱导的前列腺癌DU145及PC3细胞增殖中的作用

肝细胞生长因子(HGF)能诱导前列腺癌细胞增殖、转移及侵袭。同时,瞬时受体电位通道C6(TRPC6)与前列腺癌细胞增殖密切相关。因此我们设计试验检测TRPC6通道在HGF诱导的前列腺癌细胞增殖中的作用。应用RT¬PCR及Western blotting 检测TRPC6的表达。利用全细胞膜片钳技术及钙成像观察TRPC6通道的活性。另外,我们检测TRPC6通道对前列腺癌细胞周期、细胞凋亡及细胞增殖的影响。结果发现DU145及PC3细胞表达TRPC6及c-Met。再者,沉默TRPC6通道可抑制OAG诱导的TRPC样电流,因此DU145及PC3细胞中的存在功能性的TRPC6通道。此外,沉默TRPC6通道使DU145及PC3细胞生长停滞在G2/M期,同时抑制HGF诱导的细胞增殖。总之,TRPC6在HGF诱导的DU145及PC3细胞增殖中起重要作用。
