组织工程和RNA干扰技术用于尿道重建的初步研究

本研究目的在于探讨以口腔粘膜细胞和TGF-β1 siRNA转染的成纤维细胞为种子细胞替代尿路上皮细胞,以膀胱脱细胞基质(BAMG)为支架进行尿道重建的可行性。将TGF-β1 siRNA转染兔成纤维细胞,通过RT-PCR,检测siRNA对兔TGF-β1表达的干扰效果,把筛选出的有最佳干扰效率的siRNA,重新转染兔成纤细胞,分别取成纤维细胞培养液行ELISA检测,观察细胞培养液中合成I型胶原蛋白变化情况。然后将口腔粘膜细胞和TGF-β1 siRNA转染的成纤维细胞为种子与BAMG复合,构建组织工程化黏膜。应用扫描电镜对构建的组织工程化黏膜进行观察评估。结果发现TGF-β1 siRNA转染成纤维细胞后,其I型胶原蛋白合成明显降低。将口腔粘膜细胞和TGF-β1 siRNA转染的成纤维细胞为种子与BAMG复合,应用扫描电镜观察发现口腔粘膜细胞和TGF-β1 siRNA转染的成纤维细胞与BAMG有良好的生物相容性,生长良好。研究结果证实TGF-β1 siRNA可抑制成纤维细胞I型胶原蛋白的分泌,为尿道瘢痕的基因治疗提供了理论依据。口腔粘膜细胞和TGF-β1 siRNA转染的成纤维细胞与BAMG有良好的生物相容性,该组织工程化黏膜可作为尿道重建的一种新的选择。
