Col1a1表达下调诱导小鼠精原细胞分化

细胞外基质(ECM)在细胞的增殖和分化中具有重要作用,Col1a1是I型胶原蛋白的一个亚型,是细胞外基质的重要组成成分。但是,Col1a1在精子发生过程,尤其是在精原干细胞(SSCs)的增殖与分化中的调控作用尚不清楚。本研究中,我们探索Col1a1表达下调时对小鼠精原细胞增殖和分化的影响。研究显示当Col1a1表达下调时,SSCs的增殖能力的标志基因Oct4和Plzf的转录水平亦明显下降。与此同时,SSCs分化的标志基因c-kit 和 haprin的转录水平得到了加强。此外,当Col1a1表达下调时,细胞周期研究表明,有2/3的精原细胞阻滞在细胞周期的S期;体内实验进一步显示,质粒pSPAPI被注射入生精小管后再电转,管壁上的精原细胞数量明显减少。我们的研究提示Col1a1表达沉默时,精原细胞的分化能力得到促进,同时其自我增殖受到抑制。
