HIF1A基因单核苷酸多态性与中国人群前列腺癌发病风险的相关性研究

缺氧诱导因子-1α(hypoxia-inducible factor-1α, HIF-1α)在血管生成调节过程中起重要作用,而血管生成是肿瘤生长及转移的必要条件。有研究表明HIF-1α编码基因HIF1A的遗传变异和多种肿瘤的个体易感性相关。然而,位于HIF1A基因的单核苷酸多态性(SNPs)与前列腺癌 (PCa)发病风险的关系仍不明确。本研究应用Taqman法测定了HIF1A基因上3个潜在功能性SNPs (rs11549465, rs11549467 和 rs2057482),并采用病例-对照研究方法分析其与中国人群PCa发病风险的相关性。病例组及对照组分别包括662名PCa患者及716名健康对照者,都来自于中国汉族人群。实验结果表明,相对于rs11549467 GG基因型携带者,突变型GA+AA携带者的PCa发病风险明显增加[调整后比值比(OR)=1.70, 95%可信区间(CI)=1.06-2.72,这一现象在年老人群(OR=2.01, 95% CI=1.05-3.86)、吸烟人群(OR=2.06, 95% CI=1.07-3.99)、不饮酒人群(OR=2.16, 95% CI=1.20-3.86)以及无癌症家族史人群(OR=1.71, 95% CI=1.02-2.89)中更为明显。此外,rs11549467突变基因型携带者的Gleason评分相对较高(OR=2.14, 95% CI=1.22-3.75)。而rs11549465 和 rs2057482与中国人群PCa发病风险无明显的相关性,但是同时携带rs11549465和rs2057482突变型的个体PCa的发病风险明显增加(OR=2.10, 95% CI=1.23-3.57)。我们的研究结果表明HIF1A基因单核苷酸多态性影响中国汉族人群PCa易感性及疾病进展。
