中国人口中TP53和MDM2基因变异与男性不育

TP53是一种转录调节因子和肿瘤抑制因子,在精子的形成过程中发挥重要功能;MDM2为p53通路中调节p53活性的关键因子。这两种蛋白的功能均与生殖细胞的凋亡相关,而生殖细胞的凋亡可能影响生育。但目前这两个基因多态性对生殖细胞凋亡及男性不育影响的普遍性方面知之甚少。本研究主要探讨TP53中72Arg>Pro(rs1042522)与the Ex2+19C>T(rs2287498),MDM2中5' UTR 309T>G (rs937283)这三个点的多态性与男性先天不育的关联性。我们采用OpenArray基因分型方法,对包括580例不育病人和580例正常生育对照的病例-对照样本进行了研究。结果显示TP53 Ex2+19C>T和MDM2 309T>G的多态性与男性不育具有相关性。此外,我们采用相加模型对TP53 rs2287498与MDM2 rs937283交互作用进行了分析,结果近似具有统计学意义(P=0.055)。总之,本研究为TP53通路基因多态性与男性不育的相关性提供了初步证据。
