NKX3.1对前列腺癌PC3细胞中IGF-1R的表达及其信号转导通路的抑制作用研究

人同源盒基因NKX3.1在前列腺中特异表达与前列腺癌的发生、发展密切相关,该基因通常作为抑癌基因发挥作用。本实验旨在研究NKX3.1对前列腺癌细胞PC3中IGF-1R的表达及其信号通路的影响。NKX3.1真核表达载体pcDNA3.1-NKX3.1稳定转染PC3细胞,稳定转染空载体组作为对照。RT-PCR和Western blotting 分别检测两组细胞中IGF-1R mRNA 和蛋白质的表达;Western blotting和荧光素酶报告基因检测IGF-1/IGF-1R信号通路下游分子的表达和活性;不同浓度IGF-1处理上述两组细胞,MTT和流式细胞术检测细胞生长增殖。结果显示,NKX3.1在PC3细胞中表达后大大降低了IGF-1R mRNA和蛋白质的表达,同时抑制了IGF-1诱导的ERK1/2和AKT的磷酸化、Elk-1转录因子活性及下游靶基因c-fos和cyclinD1的表达;另外,NKX3.1表达后,显著抑制了IGF-1对细胞增殖的诱导作用。我们从中得出结论:NKX3.1能够降低PC3细胞中IGF-1R的表达,抑制IGF-1R介导的MAPK/ERK和AKT信号通路,从而抑制IGF-1诱导的细胞增殖。本研究阐明了NKX3.1抑制肿瘤的新的分子机制,并为前列腺癌的治疗提供了新的思路。
