异相睡眠剥夺和睡眠恢复对不同年龄大鼠睾酮水平的影响

本研究评估了异相睡眠剥夺(PSD)引起的血清睾酮的变化情况,并检验了睡眠恢复(SR)对PSD不同持续时间及不同年龄大鼠的血清睾酮水平的补给情况。将12周龄的Wistar雄性大鼠作为青年组(YG),20周龄的作为老龄组(EG),并随机分配到对照组(笼和平台),3、5、7天睡眠剥夺(SD)组和1、3、5天SR组中。用改良的多重平台法限制动物快速眼动(REM)睡眠制作PSD模型。评估在PSD和SR恢复期间,YG和EG组的睾酮和促黄体激素水平差异。在睡眠剥脱期,YG组(P=0.001, 相关系数=-0.651)和EG组(P=0.001, 相关系数=-0.840)的睾酮水平均呈时间依赖型下降。与YG组相比,EG组大鼠在睡眠剥夺后睾酮水平有显著性下降。3天睡眠剥夺实验后,我们紧接着对所有大鼠进行了5天的睡眠修复,YG组的睾酮水平持续上升,而在EG组中直到第5天,睾酮水平仍然没有恢复。相比YG组,PSD对EG组血清睾酮水平的伤害更大,且会伴随促黄体激素水平的下降。EG组大鼠血清中的睾酮补给水平受到了抑制。
