淫羊藿次苷Ⅱ对DM性ED大鼠阴茎勃起功能、AGEs、自噬、mTOR 和 NO-cGMP影响及其机制的研究

勃起功能障碍(ED)是糖尿病的主要并发症,并且已经被证实淫羊藿苷是通过其生物活性形式淫羊藿次苷Ⅱ发挥作用而增强勃起功能。本实验研究淫羊藿次苷Ⅱ对DM性ED大鼠阴茎勃起功能、阴茎海绵体组织中晚期糖基化终产物(AGEs)、自噬、mTOR和NO-cGMP信号通路影响及其机制。通过酶化学法从淫羊藿苷分离提纯出淫羊藿次苷Ⅱ。淫羊藿次苷Ⅱ作用糖尿病ED组大鼠灌胃淫羊藿次苷Ⅱ,而正常对照组和糖尿病ED组大鼠分别灌胃相应量的生理盐水。阴茎海绵体压力和平均动脉压比值(ICP/MAP)测定同组别大鼠阴茎勃起功能,酶联免疫法测定不同组别大鼠海绵体组织中AGEs、NOS活性和cGMP含量,四甲基偶氮唑盐微量酶反应比色方法(MTT法)和流式细胞仪法分析细胞增殖状况。透射电子显微镜法、单丹磺酰尸胺(MDC)染色法和GFP-LC3荧光自噬共定位法观察测定自噬状况,蛋白质免疫印迹法测定NOS异构体和自噬的关键蛋白的表达;并比较三组间上述指标的差异程度。在淫羊藿次苷Ⅱ作用糖尿病ED组大鼠中,淫羊藿次苷Ⅱ增加了ICP/MAP、平滑肌细胞生长曲线、S期和平滑肌细胞含量,而降低了Beclin1(P<0.05);淫羊藿次苷Ⅱ显著增加了平滑肌细胞增殖指数和p-p70S6K (Thr389)表达,显著减弱了平滑肌细胞中的自噬和LC3-II表达(P<0.01);淫羊藿次苷Ⅱ减弱了AGEs,增加cGMP含量和NOS活性(P<0.05),并且显著增加了cNOS(P<0.01)。淫羊藿次苷Ⅱ明显改善DM性ED大鼠阴茎勃起功能,这可能与淫羊藿次苷Ⅱ上调阴茎海绵体平滑肌细胞增殖和NO-cGMP信号通路及下调AGEs、自噬和mTOR信号通路有关。
