【原创论文】睾丸特异表达F a n k1基因敲减小鼠产生少精症表型涉及凋亡途径

Fank1基因是一个睾丸组织特异表达基因,在精子发生过程的减数分裂到单倍体阶段广泛表达。本文通过RNA干扰技术建立Fank1基因敲减转基因小鼠模型来分析Fank1基因功能,结果显示Fank1基因敲减小鼠的精子数量减少及精母细胞凋亡增加,Fank1基因表达缺失可导致Fank1基因直接或间接调控的多个靶基因表
达异常,引起精子生成障碍。应用CAST实验获得FANK1的DNA结合序列AAAAAG,通过基因芯片技术筛选出基因敲减小鼠与野生型小鼠表达差异基因,在它们的启动子区域均发现AAAAAG FANK1的DNA结合序列。因此Fank1是精子发生过程中重要的转录调控因子。
