α酮酸保护大鼠精子免受氧化应激损伤的体外研究

本研究旨在检测抗氧化剂包括α酮酸(α酮戊二酸、丙酮酸)、乳酸和谷氨酸/马来酸,对大鼠精子受到氧化应激伤害时的保护作用。我们的结果显示H2O2 (250 μmol L-1)造成的损伤,例如运动能力下降、三磷酸腺苷(ATP)减少、精子蛋白酪氨酸磷酸化受到抑制、顶体反应和存活率降低等能够被α酮戊二酸(4 mmol L-1)或丙酮酸(4 mmol L-1)有效阻止。当培养液中无外源H2O2时,添加丙酮酸(4 mmol L-1)能显著提高精子悬液中超氧阴离子(O2-•)的水平(P < 0.01),添加α酮戊二酸(4 mmol L-1)和乳酸(4 mmol L-1)能显著增强精子95kDa蛋白的酪氨酸磷酸化程度(P < 0.04)。同时α酮戊二酸、丙酮酸、乳酸、谷氨酸和马来酸也是精子运动所需能量和ATP的重要来源。本研究结果表明α酮酸可以作为有效的抗氧化剂保护大鼠精子免受H2O2的攻击,同时能够增强BWW(Biggers, Whitten and Whittingham)培养液的抗氧化能力。
