GnRH并列体二聚物主动免疫降低SD雄鼠血
清生殖激素浓度,抑制睾丸发育和精子发生
由于诸多实践和临床原因,GnRH免疫去势疫苗得到
大力开发和应用。然而,将GnRH分子与载体蛋白偶联以
增强抗原免疫原性的方法存在诸多不足,阻碍了GnRH去
势疫苗的商业化应用。本研究评价非载体偶联GnRH去势
抗原---GnRH并列体二聚物(D-Lys6-GnRH-tandem-dimer
peptide,TDK)配以Specol佐剂对SD雄鼠生殖功能的抑制
效果。36只青春期前期SD雄鼠随机分为3组(n = 12):
对照组(不做任何处理),手术去势组(试验开始前2周
进行手术去势)和免疫组。免疫组于6周龄时初免100 ug
TDK Specol乳化剂 (1 ml), 8周后加免。试验开始后每2周
收集血液直至处死大鼠(18 周龄)以测定血清抗体滴度
及激素浓度。结果显示,TDK主动免疫降低大鼠血清T、
抑制素B,LH及FSH浓度(P < 0.05),抑制大鼠青春期
睾丸精子发生(P < 0.05)。与对照鼠相比,TDK主动免疫
下调垂体GnRH受体、LH-β、FSH-β,及睾丸LH受体、
FSH受体、抑制素α、βA、βB 基因mRNA表达水平(P
< 0.05)。结果首次表明,TDK配以Specol佐剂有望作为
一种新型的免疫去势疫苗。
