糖尿病大鼠阴茎海绵体组织中SK3和IK1通道蛋白表达降低

中、小型钙激活性钾通道(SK3、IK1)在内皮依赖性超极化因子通路中扮演重要角色。我们检测了SK3和IK1在糖尿病大鼠阴茎组织中的表达。采用8周龄的SD雄性大鼠(250–300克)腹腔注射链脲佐菌素制作糖尿病动物模型。同时,对糖尿病组(20只)和对照组(10只)大鼠皮下注射阿扑吗啡,观察阴茎勃起情况;采用RT-PCR和Western blot分别检测SK3和IK1 mRNA和蛋白水平。结果显示:糖尿病组大鼠阴茎勃起功能明显低于对照组,差异具有统计学意义(P < 0.05)。糖尿病组大鼠海绵体中SK3和IK1表达显著低于对照组(P < 0.05)。糖尿病可抑制海绵体中SK3和IK1在mRNA和蛋白的表达。SK3和IK1表达的减少可能在糖尿病性勃起功能障碍的发病中具有重要作用。
