化学性糖尿病大鼠模型反复给予udenafil导致阴茎海绵体中一氧化氮合酶基因、蛋白表达上调

勃起功能障碍(ED)是糖尿病(DM)的一个主要并发症。本文主要研究了ED与糖尿病大鼠阴茎海绵体(CC)中结构性一氧化氮合酶(cNOS)下调的关系,检测了udenafil,一个5型磷酸二酯酶(PDE5)抑制剂,在ED和cNOS表达水平上的作用。糖尿病大鼠口服udenafil共16周,记录阴茎内压/平均动脉压(ICP/MAP)比值,应用反转录聚合酶链反应(RT-PCR)和免疫印迹法来检测cNOS表达。尽管糖尿病大鼠阴茎海绵体的ICP/MAP比值以及表皮NOS (eNOS)、神经NOS (nNOS)的表达水平显著降低,但是,与糖尿病对照比较,长期udenafil治疗可以改善勃起功能障碍,增加cNOS表达水平。这些结果显示,糖尿病ED与阴茎海绵体的cNOS表达降低密切相关,udenafil可以通过调节cNOS表达来修复病理改变,udenafil在环磷酸鸟苷(cGMP)降解中起抑制作用。
