【论著】次级淋巴趋化因子与肿瘤抗原构建树突状细胞瘤苗在小鼠前列腺癌中的研究

目的:探讨转染次级淋巴趋化因子基因与肿瘤抗原的树突状细胞瘤苗在小鼠前列腺癌中的抗肿瘤效应。方法:通过脂质体法将次级淋巴趋化因子基因转染至小鼠骨髓来源的树突状细胞,同时将RM-1前列腺癌细胞的裂解产物负载树突状细胞构建DC瘤苗;逆转录聚合酶链反应检测SLC基因成功转染;检测DC瘤苗的抗前列腺癌的免疫治疗作用。结果:与SLC-DC、Lysate-DC、DC及PBS组相比,SLC+Lysate-DC组明显抑制肿瘤生长(P<0.01)。免疫组化荧光染色可见相对于其他各组,SLC+Lysate-DC组中有更多的CD4+、CD8+T细胞和CD11+DC浸润。结论:转染SLC+Lysate的DC瘤苗能通过趋化大量CD4+、CD8+T细胞及CD11+DC诱导抗肿瘤的免疫效应,为前列腺癌的治疗提供了新的免疫治疗手段。
