【论著】评估枸橼酸西地那非处理的大鼠海绵体组织血红素加氧酶-1的活性

目的:评估枸橼酸西地那非处理的大鼠海绵体组织中血红素加氧酶-1(HO-1)的活性。方法:把192只SD雄性大鼠平均分为四组进行研究: 组1,对照组,喂食正常动物食物;组2,经胃管喂食枸橼酸西地那非;组3,喂食枸橼酸西地那非和HO抑制剂(锌原卟啉,ZnPP);组4,喂食枸橼酸西地那非和一氧化氮合成酶(NOS)抑制剂L-硝基精氨酸甲酯(L-NAME)。每组12只大鼠在给药0.5小时,1小时,2小时和3小时后处死。检测海绵体组织中HO-1活性,cGMP水平和NOS酶活性。结果:在枸橼酸西地那非处理的大鼠海绵体组织中,HO-1活性,cGMP水平和NOS酶活性比其它实验组相比明显升高。经过HO或NOS抑制剂处理的大鼠的这些参数明显降低。海绵体组织中HO-1和NOS酶活性与cGMP的水平呈正相关性(r=0.646, r=0.612; P<0.001)。结论:作为PDE5抑制剂的枸橼酸西地那非对海绵体组织的作用受到HO-1和NOS之间的相互作用的调控。
