【简报】精子DNA比血液DNA具有更高Yq微缺失的频率

目的:检测在不同胚胎来源的两种组织(血液和精子)中Yq微缺失的频率和位点是否相似。方法:本研究包括52名不育的少精症病人,提取每个病人的血液和精子来源的基因组DNA,并通过PCR方法进行微缺失分析。PCR产物用1.8%琼脂糖胶分析。阴性的PCR扩增产物至少重复三次来确定标记物的缺失。结果:在血液DNA中只有1例发生缺失,而精子DNA有4例缺失。结论:生殖细胞的Yq微缺失的频率高于血液。由于大多数不育夫妇选择辅助生殖技术(ART),因此生殖细胞中Yq微缺失的筛查对了解不育的遗传基础十分重要,可以对不育夫妇提供全面的咨询和最合适的疗法。
