NAC 通过抑制内质网应激保护镉诱导的睾丸生殖细胞凋亡

镉是一种生殖毒物,能够诱导睾丸生殖细胞凋亡。最新的研究发现内质网应激参与了镉诱导的睾丸生殖细胞凋亡。本研究采用抗氧化剂N-乙酰半胱氨酸(NAC )探讨其对镉诱导的睾丸内质网应激与生殖细胞凋亡的影响。研究结果显示NAC 预处理能明显减轻镉诱导的睾丸生殖细胞凋亡。糖调节蛋白78(GRP78 )是重要的内质网特异性分子伴侣,NAC 显著下调镉诱导的GRP78 的表达。翻译起始因子(eIF2α)是内质网应激PERK 信号通路下游的靶分子,而NAC 预处理明显降低镉诱导的eIF2α蛋白的磷酸化。此外,NAC 能阻断镉诱导的X盒结合蛋白1 (XBP1)mRNA 的激活。上述结果提示抗氧化剂NAC 减轻镉诱导的睾丸内质网应激和非折叠蛋白反应。c-Jun 氨基末端激酶(JNK )与C/EBP 同源蛋白(CHOP )是内质网应激介导的凋亡信号通路的两个重要分子,NAC 几乎完全阻断镉诱导的JNK 蛋白磷酸化与CHOP 的表达,提示NAC 预处理能够通过抑制内质网应激保护镉诱导的睾丸生殖细胞凋亡。
